EFFECT OF SALINITY ON IN VITRO PERFORMANCE OF TOMATO (Lycopersicon esculentum L.)

The present investigation was aimed at exploring the effect of Salinity on in vitro culture of tomato plant. 50mM, 100mM and 150mM of NaCl and CaCl2 as well as 40mM, 80mM and 120mM of Na2SO4 practiced in the experiment. A total of five characters were observed i.e. seed germination percentage, average seedling height (cm), average number of leaves, callus induction percentage and regeneration percentage. Significant differences were found for differences in both salt types and concentration. All the characters gradually decreased with higher salt concentration. Among different types of salt, Na2SO4 exhibited more detrimental effect on all the studied characters than NaCl and CaCl2. The highest seed germination was obtained for 50 mM NaCl whereas germination was not found in 120mM Na2SO4. For seedling height, the salinity effect was not prominent. The study also detected that NaCl and CaCl2 produced more leaves than Na2SO4. The higher dose of Na2SO4 i.e. 120mM Na2SO4 did produce any germination. Calli were not formed also for the higher doses of salt. Both NaCl and CaCl2 produced the same calli percentage @ 50mM dose. The calli failed to regenerate successfully in case of 80 mM Na2SO4. In case of regeneration, both NaCl and CaCl2 produced the same plantlet percentage in their 50mM dose. Regeneration was not observed at higher doses of salt also for 80 mM Na2SO4.
